Description:

NNMTi is an effective inhibitor of nicotinamide N-methyltransferase (NNMT) (IC50=1.2 μ M) that selectively binds to NNMT substrate binding site residues. NNMTi can promote the differentiation of muscle cells in vitro, enhance the fusion and regeneration ability of muscle stem cells in elderly mice.

Application

1. Inhibit NNMT activity: Reduce the methylation of nicotinamide (NAM) to 1-methylnicotinamide (1-MNA), thereby increasing intracellular NAD ⁺ levels, enhancing sirtuin1 (SIRT1) activity, and improving cellular metabolism.

2. Promote muscle regeneration: In an elderly mouse model, NNMTi can enhance the proliferation and fusion ability of muscle stem cells (muSCs), and improve skeletal muscle function.

3. Anti obesity effect: Inhibits NNMT expression in adipose tissue, reduces fat accumulation, and improves liver steatosis.
