As the years pass, skeletal muscle mass and strength diminish in the elderly, along with its regenerative capacity, which often leads to symptoms of muscle decline closely associated with aging and a higher incidence of disease. Muscle stem cells (abbreviated as muSCs) begin to dysfunction as they proliferate and differentiate into myoblasts with fusion potential with age, which in turn affects the ability of muscles to recover after damage.
The small molecule inhibitors developed to fill this gap target nicotinamide N-methyltransferase (High quality nnmti), an enzyme whose expression increases significantly during skeletal muscle aging, and which is strongly associated with impairment of the NAD+ salvage pathway, dysregulation of sirtuin 1 activity, and exacerbation of senescence in MSCs.
High quality nnmti on the cell membrane plays a key role in regulating the level of nicotinamide precursors required for NAD+ biosynthesis, thus playing a crucial role in the NAD+ salvage pathway and cellular metabolic regulation.
In skeletal muscle tissues, the expression of NNMT showed a positive correlation with the aging process of muscle, i.e., its expression gradually increased with age.
In particular, NNMT has become a central element in the gene expression signature of sarcopenia, and its over-activity may lead to a significant decrease in NAD+ levels, which in turn may lead to aging-related loss of muscle mass, strength, and functional deterioration.
In response to this phenomenon, small molecule inhibitors of high quality nnmti have been developed that possess drug-like chemical properties and can precisely and selectively inhibit NNMT activity in vivo.
