First, the mechanism of action of white kidney bean extract primarily targets starch in food. If someone who is dieting avoids carbohydrates, their diet typically lacks starch intake, so white kidney bean extract cannot exert its effects.

Second, carbohydrates come in various forms, such as glucose, fructose syrup, and other monosaccharides and disaccharides. The protein content in the supernatant is then measured.

2. Albumin extraction: Take white kidney bean powder with a mesh size of 60, add distilled water at a material-to-liquid ratio of 1:8 (g/mL), soak at 55°C for 3 hours, then centrifuge at 4000 rpm for 20 minutes. The supernatant is the white kidney bean albumin extraction solution.

3. Extraction of α-amylase inhibitors: There are two main methods for preparing α-amylase inhibitors: fermentation and plant extraction. For α-AI obtained from plant sources, the main steps in its separation and purification include grinding, heat treatment or water extraction, precipitation with ammonium sulfate or ethanol separation, DEAE-cellulose column chromatography, Sephadex G-75 gel chromatography, and CM-cellulose column chromatography, followed by freeze-drying to obtain a white powdery enzyme. Another reported purification method involves: soaking plant seeds in water → grinding the tissue → centrifugation to collect the supernatant → rapid heating for selective thermal denaturation → cooling to room temperature, followed by centrifugation to collect the supernatant → salt precipitation → buffer dialysis → DEAE-32 chromatography → polyethylene glycol 6000 concentration → affinity chromatography → concentration and freeze-drying.
